M). The cultures were incubated at 37° for 40 mm (“Pretreatment―), whereupon the bacteria were washed and resuspended in fresh Medium HA. Portions from each culture were distributed into flasks containing premeasured amounts of the various drugs (“Additions―). After receiving cytidine-3H (2 @ig/ml; 0.13 i@Ci/ml),the cultures were incubated with aeration at 37°for 20 mm and quadr.uplicate 0.5-mi portions were withdrawn from each culture; 2 of these were processed immediately for radioactivity determination (“Total nucleic acids―), while the other 2 were subjected to alkali digestion (18) prior to radioactivity measurement (DNA).

متن کاملEffect of hydroxyurea on replication of bacteriophage T4 in Escherichia coli.
Hydroxyurea inhibited the replication of bacteriophage T4 in Escherichia coli B. The concentration of hydroxyurea required to inhibit net deoxyribonucleic acid (DNA) synthesis 50% was about 50-fold less than that required in uninfected cells. Even in the presence of high hydroxyurea concentrations, phage DNA was readily synthesized from the products of breakdown of the E. coli DNA, and viable p...
متن کاملOxygen-sensitive ribonucleoside triphosphate reductase is present in anaerobic Escherichia coli.
Ribonucleoside diphosphate reductase from Escherichia coli and mammalian cells provides the deoxyribonucleoside triphosphates for DNA synthesis. The active enzyme contains a tyrosyl free radical whose formation requires oxygen. Earlier genetic evidence suggested that the enzyme is not required for anaerobic growth of E. coli, implicating the activity of a different enzyme or enzyme system for d...

The preceding paper (1) described the purification from Escherichia coli of thioredoxin, a heat-stable, small protein involved in the reduction of cytidine diphosphate to deoxycytidine diphosphate by the cytidine diphosphate reductase system isolated from E. coli (2). Evidence was presented that thioredoxin was first reduced enzymatically with TPNH to a sulfhydryl form (thioredoxin-(SH)J, which...

Escherichia coli class Ib ribonucleotide reductase (RNR) converts nucleoside 5'-diphosphates to deoxynucleoside 5'-diphosphates in iron-limited and oxidative stress conditions. We have recently demonstrated in vitro that this RNR is active with both diferric-tyrosyl radical (Fe(III)(2)-Y(•)) and dimanganese(III)-Y(•) (Mn(III)(2)-Y(•)) cofactors in the β2 subunit, NrdF [Cotruvo, J.
